Purpose: To investigate the link between obesity and the -243A>G polymorphism in the glutamate decarboxylase 2 GAD2 gene. Method: This study involved 20 obese participants (which constituted the study group) from Al Azhar University Hospital, recruited between January 2024 and January 2025, and a control group of 20 healthy participants. -243A>G GAD2 polymorphism was identified in the peripheral blood of both study and control groups using polymerase chain reaction-restriction fragment length polymorphism (PCR RFLP) technique. Result: This study found that control group had significantly higher frequencies of the AA genotype and “A” allele compared to study group (p < 0.01). Furthermore, frequencies of the AG and GG genotypes, in addition to the “G” allele, were significantly higher in study group compared to control group (p < 0.01). Conclusion: The study revealed a significant link between the -243A>G polymorphism and obesity.
